N-terminal amino acid sequences are determined by Edman degradation with phenylthioisocyanate using the Procise Protein Sequencing System (Applied Biosystems, 491 HT or 494 cLC Protein Sequencer). The resulting PTH-amino acids are analyzed sequentially by reversed-phase HPLC, which yields the amino acid sequence of the N-terminus of the protein or peptide.

Samples can be: liquid, reblotted to PVDF, on resin (synthetic peptides). Limit of detection is 1 picomol.
